We are taking our 3 piggies for their first wellness check. What do you put them in for transport? We use fleece at home so I thought perhaps a cardboard box with fleece on bottom and some hay inside?

What about water it should be around 2 hours we are gone from their cage.
 
I use a foot and a half tall storage container with a hole cut in the lid. Mine have been without water for that long, you can offer some cucumber.
 
When travelling you require something study and secure for their safety. Please never use a cardboard box - it would be very unsafe and would offer no protection for them in the event of an accident or the box being dropped (plus they could chew out).

Please use a cat carrier. You will require a large one given you need to put all three in at once. Put something absorbent in the bottom like a towel and then add some hay and some cucumber.
If it gets cold in winter in your area, please ensure you put a blanket over the carrier and ensure they are kept warm inside (perhaps even offer a microwaveable snugglesafe) - for indoor piggies, going outside and being exposed to a sudden temperature shift (either up or down) is risky for them.

Travelling with guinea pigs
 
I take my piggies to the vet in a large cat carrier. I put a large pee-pad (or two smaller ones) in it to cover the base, but have also used a folded bath mat in the past. As long as it gives them some grip and absorbs pee. If you have nervous piggies you can use a pee pad straight from their cage so it has a strong smell of home. Then put in a handful of hay so the piggies have something to nibble on and to hide in.
I put the carrier at the footwell of the passenger seat in the car and cover it with a large towel to both make them feel safe and to reduce airflow from the heating or A/C. And then push up the passenger seat to the carrier so it doesn't move if I have to break suddenly.

They will be fine for 2 hours without water, but if they are used to it you can offer them some cucumber to keep them hydrated.
 
I use a cat/rabbit carrier with a fleece blanket on the bottom. I give them some hay and treats to keep them occupied while they're in there.
